Protein Sensors for Membrane Sterols

Cholesterol is an essential component of animal cell membranes, and its concentration is tightly controlled by a feedback system that operates at transcriptional and posttranscriptional levels. Here, we discuss recent advances that explain how cells employ an ensemble of membrane-embedded proteins to monitor sterol concentrations and adjust sterol synthesis and uptake.

Functional convergence of hopanoids and sterols in membrane ordering.

Liquid-ordered phases are one of two biochemically active membrane states, which until now were thought to be a unique consequence of the interactions between eukaryotic membrane lipids. The formation of a liquid-ordered phase depends crucially on the ordering properties of sterols. However, it is not known whether this capacity exists in organisms that lack sterols, such as bacteria. We show t...

Membrane sterols and the development of the preimplantation mouse embryo.

The role of membrane sterols in the compaction and subsequent development of the preimplantation mouse embryo was studied by incubating embryos in 7-ketocholesterol and other oxygenated sterols. These sterols have been shown to inhibit sterol synthesis and deplete membranes of cholesterol in a variety of ther cell types. Compaction and subsequent blastocyst formation were normal when embryos we...

Transport of sterols to the plasma membrane of leek seedlings

To investigate the intracellular transport of sterols in etiolated leek (Allium porrum L.) seedlings, in vivo pulse-chase experiments with [1-14C]acetate were performed. Then, endoplasmic reticulum-, Golgi-, and plasma membrane (PM)-enriched fractions were prepared and analyzed for the radioactivity incorporated into free sterols. In leek seedlings sterols are present as a mixture in which (24R...

Delivery of liposome membrane-associated sterols through silastic membranes.

The transport of sterols incorporated into the lecithin bilayer of small unilamellar liposomes through a model membrane was studied. A two-chamber diffusion cell containing liposomes with incorporated [4-14C)cholesterol or beta-[4-14C]sitosterol in the donor chamber and liposomes with unlabeled cholesterol in the receiver chamber was used. The permeability coefficients of the sterols through si...
